If you want to get all Freudian, I'd say it has to do with the difference between how a man and woman have sex and two women do.

From my understanding, not all human men have the same sex organs, and neither do all women. If it were a matter of penetration, he'd have to do more than specify what gender you're prohibited from having sex with.
